Contexte de travail/Working context

Unité mixte de recherche du CNRS, de l’INSERM et d’Aix-Marseille Université, le CIML est composé d’environ 200 personnes et 16 équipes de recherche et situé sur le parc scientifique et technologique de Luminy à Marseille (France). Au sein du CIML, l’étudiant(e) recruté(e) travaillera dans l’équipe de recherche « Cellules dendritiques et immunité antivirale » dirigée par Marc Dalod, et plus précisément dans le groupe dirigé par Elena Tomasello. L’étudiant(e) recruté(e) travaillera en étroite collaboration avec une IE recrutée sur le même projet et avec la plateforme de génomique du CIML dirigée par Pierre Milpied.

The CIML is a joint research unit of the CNRS, INSERM and Aix-Marseille University. It is composed of about 200 people and 16 research teams and is located in the Luminy Science and Technology Park in Marseille (France). Within the CIML, the student will work in the research team « Dendritic cells and antiviral immunity » led by Marc Dalod, and more precisely in the group led by Elena Tomasello. The recruited student will interact closely with an engineer recruited on the same research project and with the genomic facility of the CIML directed by Pierre Milpied.

 

Missions

L’étudiant(e) recruté(e) étudiera les mécanismes moléculaires et cellulaires régissant l’hétérogénéité fonctionnelle des cellules dendritiques plasmacytoïdes (pDC) résidant dans différents organes.

Lors d’infections virales systémiques, les cellules dendritiques plasmacytoïdes (pDC) promeuvent l’immunité antivirale via la production d’interférons de type I/III (IFN). Cette fonction est préférentiellement exercée par les pDC de la rate (Zucchini et al. Int Immunol 2008), alors que les pDC du foie et de l’intestin induisent la tolérance (Reizis, Immunity 2019). Le premier objectif sera de déterminer si des niches cellulaires spécifiques à ces trois organes instruisent des fonctions distinctes dans les pDC y résidant. Au cours de l’infection in vivo par le cytomegalovirus murin, les pDC de la rate produisant les IFN acquièrent séquentiellement 5 états d’activation ayant des fonctions et des localisations microanatomiques différentes (Abbas et al. Nat Immunol 2020). Le deuxième objectif sera de déterminer si des niches spécifiquement induites par l’infection par le MCMV régulent la génération de différents états d’activation des pDC. Ces études seront effectuées en utilisant une approche de biologie systémique, en associant de nouvelles souris mutantes ciblant spécifiquement les pDC à des technologies de pointe de génomique et d’analyse bioinformatique (Guilliams et al, Cell 2022).

The student will study the molecular and cellular mechanisms governing the functional heterogeneity of plasmacytoid dendritic cells (pDC) residing in different organs.

During systemic viral infections, plasmacytoid dendritic cells (pDCs) promote antiviral immunity via the production of type I/III interferons (IFN). This function is mainly exerted by splenic pDCs (Zucchini et al. Int Immunol 2008), whereas liver and intestine pDCs induce tolerance (Reizis, Immunity 2019). The first aim will be to determine whether organ-specific cell niches instruct distinct functions in the pDCs. During in vivo infection with murine cytomegalovirus, IFN-producing pDCs in the spleen sequentially acquire 5 activation states with different functions and microanatomical locations (Abbas et al. Nat Immunol 2020). The second aim will be to determine whether niches specifically induced by MCMV infection regulate the generation of different pDC activation states. These studies will be performed by using a systems biology approach, combining novel mutant mice specifically targeting pDCs with state-of-the-art genomics technologies and bioinformatics analyses (Guilliams et al, Cell 2022).

 

Activités/Activities

  • Isolation de différents organes de souris, préparation de sections de ces organes, marquages en immunofluorescence afin d’effectuer des expériences de « spatial transcriptomics »
  • Séquençage ARN au niveau de cellule ou du noyau unique
  • Préparation d’échantillons, marquage et acquisition par cytométrie en flux multiparamétrique
  • Préparation d’échantillons, marquage et acquisition par microscopie confocale spectrale multiparamétrique
  • Expérimentation animale (manipulation de souris vigiles, injection intraveineuse, injection intrapéritonéale)
  • Elevage et génotypage par PCR de différentes lignées de souris requises pour les expériences du projet de recherche

 

1)   Isolation of different mouse organs, preparation of sections of these organs, immunofluorescent labelling to perform “spatial transcriptomics” experiments

2)   RNA sequencing at the single cell or single nucleus level

3)   Sample preparation, labelling and acquisition by multiparametric flow cytometry

4)   Sample preparation, labelling and acquisition by multiparameter confocal spectral microscopy

5)   Animal experimentation (mouse handling, intravenous injection, intraperitoneal injection)

6)   Breeding and PCR genotyping of different mouse lines required for the research project experiments

 

Compétences/ Skills

  • Excellent niveau de connaissance en Immunologie
  • Très bonne maitrise des techniques d’histologie et de microscopie, de préférence confocale, et/ou de la cytométrie en flux (fortement souhaitée)
  • Expérience prouvée en biologie moléculaire : extraction ADN, ARN, PCR, qPCR (fortement souhaitée)
  • Expérience acquise dans le soin aux souris et leur utilisation pour l’expérimentation animale (niveau praticien/opérateur acquis, de préférence)
  • Bonne maitrise de la langue anglaise, présentation en anglais de ses propres résultats au cours des réunions d’équipe
  • Gestion des lignées de souris, génotypage

1)   Excellent level of knowledge of Immunology

2)   Very good knowledge of histology and microscopy techniques, preferably confocal, and/or of flow cytometry (strongly wished)

3)   Proven experience in molecular biology: DNA and RNA extraction, PCR, qPCR (strongly wished)

4)   Experience in care and use of mice for experimentation (practitioner/operator level wished)

5)   Good knowledge of the English language, presentation in English of own results during team meetings

6)   Management of mouse lines, genotyping
